Can you remove the internal charging cable thats attached to the usb?

The internal charging cable is very secured. It is possible to remove it if you need to replace it. If the cable is not damaged i would just leave it be.

Do I have to charge the backpack first before use?

The backpack itself does not charge. It does have a plug so you can plug in a power bank. I hope that helps.
