Is this compatible with the main villainous game or only the marvel edition?

The gameplay while similar is not the same. I have been able to adapt a few rules but gameplay just doesn't mesh completely. So, the short answer is no.

Does this allow for more players to play at one time if combined with the original?

Yes if you have the original box set which I believe allows 6 to play and 2 expansions 12 people can play and if you get more expansions because I think there are 4 expansions more people can play
